Description
Conditionally Immortalized Human Bone Marrow-Derived Mesenchymal Stem Cells (CIHMSCs) are generated from primary human BM-MSCs via lentiviral transduction with
a TetON-inducible SV40 Large T antigen (SV40 LT) expression system.
The cells exhibit typical MSC morphology: spindle-shaped, fibroblast-like cells. Proliferation is controlled by doxycycline; in the presence of doxycycline, SV40 LT is expressed, driving proliferation. Upon doxycycline withdrawal, SV40 LT expression ceases, and cells return to a quiescent, primary-like state.
Cells were selected with puromycin to ensure stable integration and expression of the transgene. These cells retain key MSC surface marker expression and multilineage differentiation potential under non-permissive conditions.
